This is not something that every search committee does, but is a way to reduce the short-list of candidates brought out for an on-campus interview.

You will be expected to give a 30-60 minute seminar about your work. Some rules of thumb gleaned from the many science blogs and other advice.
Stay focused. You can't cover everything you've done in grad school and your postdoc.
Keep it to 35-40 minutes long so there is ample time for questions, running over or to the end of time is a BAD thing.
Keep it simple. Remember this is a general science audience not your usual buddies from the specialized scientific meetings you go to, so make your audience feel smart while still covering your work.
Paint a broad picture of the context of your work and how that will transition to future work in your own lab.
Go to job talks at your institution. Note what works and what doesn't.
